Gelatine is hydrolysed by gelatinase into polypeptides and then into smaller amino acid molecules. The tubes, once inoculated, are incubated which causes the gelatine to melt. The tubes must then be refrigerated to solidify the contents before then removing and leaving at room temperate for 1 hour. If the contents are then a liquid the result is said to be positive.
